Effect of Usage of the Proton Pump Inhibitor Omeprazole on the Structure of the Kidney of Male Albino Rats

Abstract Introduction Recent clinical observational studies claimed that long term use of proton pump inhibitors (PPIs) might lead to kidney injury progress end-stage renal disease. Aim the work Was verify these claims by an in vivo experimental study rat. Materials &methods Fourty two adult male albino rats were assigned four groups Control group (I) which not administrated any treatment . In IIa, IIb, IIc received daily oral omeprazole dose 0.75mg per kg. for 2,4,or 6 weeks successively. At end experiment, blood samples collected serum creatinine and urea nitrogen (BUN) measurement. Then animals sacrificed specimens processed into paraffin blocks, sectioned stained with H&E, Mallory trichrome &PAS. Stained sections image analysis used count vacuolated cells, pyknotic nuclei, tubular casts area precent collagen fiber deposition then data was subjected statistical analysis. Results Histological examination from treated showed sectional corpuscle, tubules sever vascular congestion inflammatory cell infiltrate interstitium Thickening basement membrane seen PAS, while progressive increase detected stain Morphometry, confirmed histological findings. Statistical significant number hyaline percent compared control group. Clear deterioration function tests observed across 3 time points highly rise 4 & control. -dependent worst damage week (IV). Conclusion The present declared resulted structural rat tissue associated a dependent manner. Extreme caution should be taken prescription close monitoring is recommended.
